The majority of the new cases are work permit holders residing in foreign worker dormitories, the Ministry of Health (MOH) said in its preliminary release of figures.
Among the new cases, 15 are Singaporeans and permanent residents, while foreign workers living in dormitories continue to drive the increase.
The new cases bring the national total to 10,141.
Mr Lee also announced that circuit breaker measures to stem the spread of the coronavirus will be extended to June 1, with existing rules further tightened for the next two weeks.This means more workplaces will be closed, reducing the number of workers who keep essential services going.
